When an enemy character attacks you, a silver disc will appear under your character. Hitting the A button when this disc appears will block their attack and reduce the damage you take. Doing this 100 times will unlock this achievement. You should be blocking whenever possible anyway, so this should unlock naturally over the course of the game.
